PAPANICOLAOU STAINING SOLUTIONS
(EA 36, EA 50, EA 65, and OG 6)

RECOMMENDED PROCEDURE:   Smears are alcohol-ether fixed. Smears are brought to distilled water through alcohols. Stain 2 - 3 minutes in Harris’ or Mayer’s Hematoxylin. Place in distilled water for 10 seconds. Rinse gently to prevent cells from being washed off (about 5 to 6 dips). Wash under running tap water for 6 minutes. Rinse in distilled water and transfer through graded alcohols; 10 seconds with dipping agitation in  50%, 70%, 80% and 95% alcohol. Stain in OG 6 solution for 1.5 minutes. Rinse smear dipping for 10 seconds in two individual changes of ethanol. If mucus is present, stain 1.5 minutes in    EA 36, EA 50 or EA 65. In each of 3 baths of 95% alcohol, rinse for 10 seconds. In each  of 2 baths of absolute ethyl alcohol, rinse for 10 seconds. Clear in 3 - 4 changes of xylene.

NOTE:  For smears other than cervical scrapings, e.g. bronchial aspirates, sputum, pleural and peritoneal fluids, EA 65 is substituted for EA 50 or EA 36.

RESULTS:  Following the complete Papanicolaou method of staining, results are as indicated: nuclei stain blue with clear, sharp detail; cytoplasm stain varying shades of pink, blue, yellow, green, and gray.

SHORR-SINGLE DIFFERENTIAL STAIN

A stain for vaginal smears which provides a sharp differentiation between cornified and non-cornified elements.

RECOMMENDED PROCEDURE:   Fix wet specimen in equal parts of ethanol and ether. Stain for 1 minute. Carry through 70%, 95% and absolute alcohol - 10 dips in each solution. Clear in xylene and mount.

RESULTS:   Cornified elements stain a brilliant orange-red while the non-cornified elements take a green stain which is deeper in younger cells and paler in those more advanced. The staining is delicate and clearly reveals cytoplasmic and nuclear detail. WBC’s, RBC’s, bacteria and sperm are well-differentiated.

JENNER-GIEMSA STAIN KIT

Solution I - Jenner Stain (Stock)

Designed for staining nuclei, cytoplasm and bacteria in tissue sections.

RECOMMENDED PROCEDURE:  Working Jenner Solution:  Mix 25 ml of Sol. I with 25 ml of distilled water.   Working Giemsa Solution:  Mix 50 drops of Sol. II with 50 ml. of distilled water.  NOTE:  This solution must be made fresh. Do not re-use. Prior to staining, specimen should be well-fixed, deparaffinized and cleared. Rinse in distilled water, 2 changes, then methanol, 2 changes at 3 minutes each. Stain in working Sol. I for 6 minutes. Stain in working Sol. II for 45 minutes. Handle each slide individually in this and subsequent steps. Differentiate in 1% glacial acetic acid, then check microscopically. Rinse in distilled water. Dehydrate in 95% ethanol, absolute ethanol and clear with xylene, 2 changes each.

RESULTS:  Nuclei-blue; cytoplasm-pink to rose; bacteria-blue.

GILL-HEMATOXYLIN STAIN KIT

Gill-Hematoxylin, 0.2% (Modified), 0.4% or 0.6%
Solution II - Scott’s Tap Water Substitute
Solution III - Ethanol (95%)

An excellent nuclear stain employing a half-oxidized hematoxylin solution. (Gill, 1974) Gill Hematoxylin is a high contrast stain and a progressive hematoxylin which enchances the use of counterstains. To achieve desired stain characteristics in different specimens, the stain is available in 3 concentrations at 0.2% w/v, (Modified), 0.4% w/v, and 0.6% w/v. The modified solution at 0.2% may be preferred in cytology, however, by increasing the time (10 minutes) it can be used in histology. Gill-Hematoxylin at 0.4% is recommended for histology, however, a shorter stain time renders this solution suitable for cytologic use. For comparable results to those obtained with Harris’ Hematoxylin, Gill-Hematoxylin at 0.6% is recommended. An essential part of this stain sequence is the use of a blueing agent. We recommend Scott’s Tap Water Substitute which ensures a controlled pH and aids in cell retention on slides.

RECOMMENDED PROCEDURE:  Gill-Hematoxylin (Modified): Fix in Sol. III for 15 minutes. Water - 2 x (10 dips each). Gill-Hematoxylin (Modified) - 2 minutes. Water - 2 x (10 dips each). Sol. II - 1 minute. Water - 2 x (10 dips each). Sol. III - 2 x (10 dips each). Continue with preferred staining method.  Gill-Hematoxylin 0.4 % or 0.6 %: Xylene - 3 x  (2 minutes each). Absolute Ethanol - 3 x (30 seconds each). Water - 2 x (10 dips each). Gill Hematoxylin 0.4% or 0.6% - 3 minutes. Water - 2 x (10 dips each). Sol. II - 2 minutes. Water - 2 x (10 dips each). Continue with preferred staining method.

RESULTS:  Nuclear chromatin stains blue to blue-black; cytoplasm faintly tinted.

           MALLORY’S ACID HEMATOXYLIN STAIN
(PHOSPHOTUNGSTIC ACID)

A routine stain method for the nervous system because of its ability to demonstrate abnormal or diseased astrocytes. Useful in showing mitochondria, striations of cardiac and voluntary muscle, and as a counterstain for the Periodic Acid-Schiff (PAS) technique. Recommended staining time is 12 - 24 hours.

RESULTS:  Nuclei, mitochondria, fibrin, fibroglia, microglia, and the contractile portions of striated (voluntary) and heart muscle stain blue. Collagen, reticulum, elastin, the ground substances of cartilage and bone stain varying shades of yellow to brownish-red.

HEIDENHAIN’S IRON HEMATOXYLIN STAIN KIT

Solution I - Ammonio-Ferric Alum
Solution II - 0.5% Aqueous Hematoxylin

A histologic stain composed of ferric salts which ripen and oxidize hematoxylin rapidly, eliminating artificial or natural ripening. Solutions made with iron salts are useful for dense regressive staining. Differentiation can be accurately controlled, and the staining is consistent.

Working Solution:   Dilute Sol. II with an equal quantity of distilled water. Recommended staining time is 1 - 36 hours followed by differentiation in Sol. I.

RESULTS:  Chromatin, nucleoli, mitochondria, centrioles and certain parts of striated muscle fibers are stained black. Other tissue elements are colored by the contrast stain used.

GOMORI’S TRICHROME STAIN
(HISTOLOGICAL STAIN)

This stain demonstrates fine histological detail of connective tissue, collagen and muscle fibers.

RECOMMENDED PROCEDURE:   Deparaffinize and hydrate to distilled water. Place in Bouin’s solution in oven at 560C for 1 hour. Wash well in running water or until yellow color disappears. Stain nuclei with Weigert’s Iron Hematoxylin for 10 minutes, wash in water, then apply stain for 15 - 20 minutes. Place in 0.5% acetic water for 2 minutes. Dehydrate in 95% ethanol, absolute ethanol, and clear in xylene, 2 changes each.

RESULTS:  Muscle fibers - red; collagen - green; nuclei -blue to black.

GOMORI’S METHOD FOR IRON IN TISSUE WITH NUCLEAR FAST RED OR SAFRANIN

Designed to demonstrate deposits of hemosiderin (iron) in tissue specimens.

RECOMMENDED PROCEDURE: Working Solution:  Mix just prior to use, 50 ml of Sol. I with 50 ml of Sol. II. Run control slide. Deparaffinize and hydrate to distilled or deionized water. Cover with working solution for 20 - 30 minutes. Rinse in distilled or deionized water, counterstain with Sol. III or IIIB for 3 - 5 minutes, and rinse again. Dehydrate in 95% ethanol, absolute ethanol - 2 changes, clear in xylene - 2 changes, and mount.           NOTE: Chemically clean glassware and plastic-tipped or paraffin-coated forceps are suggested.

RESULTS: Iron pigments - bright blue; nuclei - red; cytoplasm - light pink.

Stain in Sol. III - 1 minute and rinse again. Dehydrate and clear in xylene, 2 changes each.  NOTE:  Carminophilic properties will be obscured if overstained with Working Sol. I.

RESULTS:  Capsule of Cryptococcus and mucin stain deep rose to red; nuclei stain black.

RECOMMENDED PROCEDURE:  Working Solution:  In the following order, mix 20 ml of Sol. I + 8 ml of Sol. II + 8 ml of Sol. III. NOTE:  This staining solution should,be used within 24 hours. Immerse sections in working solution for 15 minutes to 1 hour until perfectly black. Differentiate in Sol. IV for a few minutes. Examine sections in water under low magnification to observe the stages in differentiation. If the differentiation has been carried too far, (provided they have not been treated with alcohol) the sections may be restained. Wash in water. Place in 95% alcohol to remove iodine and leave sections in tap water       5 minutes or longer. Counterstain 3 - 4 minutes in Sol. V. Clear in xylene and mount.

RESULTS:  Elastic fibers - intensely blue-black; nuclei - blue to black; collagen - red.

MALLORY’S ANILINE BLUE COLLAGEN STAIN KIT

Solution I - Aniline Blue

Demonstrates collagenous fibrils, cartilage, bone, amyloid, and nuclei.

RECOMMENDED PROCEDURE:  Sections should be Zenker-fixed and procedure for removal of pigments and precipitates should be followed prior to staining. Stain with       Sol. II for 1 - 5 minutes or longer. (Omit Sol. II to highlight collagenous fibrils). Transfer directly to Sol. I without washing in water. Stain for 20 - 60 minutes or longer. Dehydrate, clear and mount. For celloidin sections, shorten staining time, decolorize, dehydrate in 95% ethanol and clear by the blotting paper xylol method or in terpineol and mount in balsam.

RESULTS:  Ground substances of cartilage and bone, mucus, amyloid - varying shades of blue; nuclei, fibroglia, myoglia and neuroglia fibrils, axis cylinders and fibrin -red; erythrocytes and myelin - yellow; elastic fibrils - pale pink, pale yellow or unstained.

 METHYL GREEN WITH PYRONIN STAIN

Unna Pappenheim Method modified by Saathof for bacteria, DNA and RNA, and plasma cells.

RECOMMENDED PROCEDURE:  Bacteria: Fixative - absolute alcohol or Carnoy’s fluid. Stain 2 - 4 minutes (up to 30 minutes if necessary). Wash in distilled water. Dehydrate quickly in absolute alcohol or acetone. Clear in xylol and mount in balsam. Celloidin sections are cleared in terpineol and mounted in balsam.

RESULTS:  Bacteria - bright red; chromatin - green; all other structures stain varying shades of red.

RECOMMENDED PROCEDURE: DNA and RNA:  Fixative - Clarke and Camoy’s fluid. Stain 10 minutes. Rinse twice in distilled water briefly. Blot with smooth filter paper.

Differentiation Solution:  Mix 30 ml Tertiary Butyl Alcohol with 10 ml 100% Alcohol. Dehydrate in a fresh change of the differentiating solution for 1 - 2 minutes. (Slides may be left in solution for 4 - 5 minutes) Clear in two changes of xylene for 10 minutes each.

RESULTS:  DNA - blue green; RNA - bright red; remainder of tissue - pale pink.

RECOMMENDED PROCEDURE:  Plasma cells:  Fixative - absolute alcohol or Camoy’s fluid. Stain 20 minutes. Rinse rapidly in water. Differentiate and dehydrate rapidly in absolute alcohol. Clear in xylol.

RESULTS:  Nuclei - green; cytoplasm - deep red.
